The Truffle can be relocated to a different house as long as the new house is also in a Glowing Mushroom biome. Trying to move him to an invalid house will give the error message, "This housing does not meet the requirements for a Truffle".
The Truffle can stay in a house after the surface Glowing Mushroom biome is destroyed. This can be exploited by creating a Glowing Mushroom biome near all of the other houses and NPCs, having him moved in, then destroying the corresponding Glowing Mushroom biome blocks, therefore having all of your NPCs in one place outside of a Glowing Mushroom biome.
This method doesn't work to relocate him underground, you need to make him a house near a mushroom biome in the surface then destroy it and leave him find an other house for that. Additionally, if the Truffle dies, this must be redone.
The Truffle is male and has a chance to be teleported by a King Statue.
Despite his requirement to live in a surface Glowing Mushroom biome, the Truffle can sell other pylons due to hybrid biomes. The exception to this are the Forest and Cavern pylons, which are mutually exclusive and only available when not within one of the other biomes.
The Truffle has arguably the second-most powerful attack of all the NPC's, beaten out only by the Princess.

The quote "As if living underground wasn't bad enough, jerks like you come in while I'm sleeping and steal my children." is a reference to players exploring Glowing Mushroom biomes and collecting Glowing Mushrooms.
The quote "I don't know the 'Truffle Shuffle,' so stop asking!" is a reference to the movie The Goonies.
The quote "I tried to lick myself the other day to see what the big deal was, everything started glowing blue." references the use of mushrooms as a recreational drug.
The quote "Everyone in this town feels a bit off. I woke up to the clothier chewing on my foot last night." implies the Clothier wants to eat the Truffle, further supported by "<Name of Clothier> has tried to eat me so many times. I swear, one time they weren't even human! I, obviously, dislike it."
This also hints the Truffle's awareness of the Clothier's former possession by Skeletron.
The Bestiary entry for the Truffle: "Harnessing the hidden power of the glowing mushroom, the Truffle forges weapons and tools exclusively in his mushroom home."
